Picornavirus 3C – a protease ensuring virus replication and subverting host responses

چکیده

ABSTRACT The protease 3C is encoded by all known picornaviruses, and the structural features related to its RNA-binding activities are conserved; these contribute cleavage of viral polyproteins assembly RNA replication complex during virus replication. Furthermore, performs functions in host cell through interaction with proteins. For instance, has been shown selectively ‘hijack’ factors involved gene expression, promoting picornavirus replication, inactivate key innate immunity signaling pathways, inhibiting production interferon inflammatory cytokines. Importantly, maintains infection subtly subverting death modifying critical molecules organelles. This Review focuses on molecular mechanisms which mediates physiological processes virus–host interaction, thus highlighting picornavirus-mediated pathogenesis caused 3C.

Recent development of 3C and 3CL protease inhibitors for anti-coronavirus and anti-picornavirus drug discovery.

SARS-CoV (severe acute respiratory syndrome-associated coronavirus) caused infection of ~8000 people and death of ~800 patients around the world during the 2003 outbreak. In addition, picornaviruses such as enterovirus, coxsackievirus and rhinovirus also can cause life-threatening diseases. Replication of picornaviruses and coronaviruses requires 3Cpro (3C protease) and 3CLpro (3C-like protease...

The Hypervariable Amino-Terminus of P1 Protease Modulates Potyviral Replication and Host Defense Responses

The replication of many RNA viruses involves the translation of polyproteins, whose processing by endopeptidases is a critical step for the release of functional subunits. P1 is the first protease encoded in plant potyvirus genomes; once activated by an as-yet-unknown host factor, it acts in cis on its own C-terminal end, hydrolyzing the P1-HCPro junction. Earlier research suggests that P1 coop...

Selective inhibitors of picornavirus replication.

Picornaviruses cover a large family of pathogens that have a major impact on human but also on veterinary health. Although most infections in man subside mildly or asymptomatically, picornaviruses can also be responsible for severe, potentially life-threatening disease. To date, no therapy has been approved for the treatment of picornavirus infections. However, efforts to develop an antiviral t...

Coxsackievirus B3 protease 3C induces cell death in eukaryotic cells

Abstract: Coxsackievirus B3 (CVB3) is the most common agent known to cause viral myocarditis. The viral genome encodes a single polyprotein that is cleaved to produce several proteins by virally encoded proteases. Most of this proteolytic processing is catalyzed by a cysteine protease called 3C. The 3C protease plays major role in viral replication and cellular damage. To understand the mecha...

Identification and characterization of a 3C-like protease from rabbit hemorrhagic disease virus, a calicivirus.

Expression studies conducted in vitro and in Escherichia coli led to the identification of a protease from rabbit hemorrhagic disease virus (RHDV). The gene coding for this protease was found to be located in the central part of the genome preceding the putative RNA polymerase gene. It was demonstrated that the protease specifically cuts RHDV polyprotein substrates both in cis and in trans. Sit...
